INFLUENCE OF CAUCASIAN RHODODENDRON (RHODODENDRON CAUCASICUM PALL.) LEAF EXTRACT ON DIABETES-INDUCED PREGNANCY LOSS

Diabetes type 1 and type 2 in pregnancy results in increased risks to both the mother and child. Beside the high rate of spontaneous abortions, babies are at increased risk of congenital malformations, perinatal mortality, macrosomia and related complications, which is thought to be attributable to poor glycemic control.

The aim of our current study is to evaluate whether oral administration of rhododendron leaf extract gives us possibility to avoid or reduce undesirable outcome by modulating mechanisms underlying diabetes-induced pregnancy loss. To clarify this question, we performed the study in streptozotocin (STZ) treated FVB female mice. To investigate influence of RhE administration on pregnancy loss in diabetic mice depending on severity of diabetes, mice daily were intook 66,67 mg/kg RhE two weeks before crossbreeding and during implantation period. Therefore, pregnancy rate and resorptions quantity were investigated on day 9 of pregnancy, when implantation period is completed. 

To characterize influence of RhE administration on expression of IL-10, TNFα, and NO in the uterine cells of diabetic mice, we analyzeduterine lysates of diabetic mice collected on days 5 and 7 of pregnancy, encompassing the period of implantation. In conclusion, the data presented in this article suggest that IL-10 and TNFα play a central role in the pathogenesis of diabetes-induced pregnancy loss. Also, RhE oral administration reduces nitric oxide generation in uterus, but do not normalize pro- and anti-inflammatory cytokine production. Although, Rhododendron Extract had a strong inhibitory effect on NO secretion but cannot regulate the mechanisms determining the outcome of diabetes-induced embryopathic stress.

The article discusses the possibility of applying Camelyn drug for correction of immune homeostasis in patients with cancer. Effect of tissue accumulation allows mountain honey peptides (MHP), which are contained in Kamelin to act as markers of mimicking cancer cells and to attract T-cells for their disintegration. The article also coveres the ability of exogenous peptides to assist in the phagocytes capture and disposal of toxic fragments of destroyed atypical cells, which is important in eliminating the consequences of chemotherapy.

IMMUNOCHISTOCHEMICAL CHANGES IN RAT PLACENTA DURING DIFFERENT PERIODS OF PREECLAMPSIA

The aim of the study was to evaluate immunochistochemical changes in different periods of preeclampsia. Placenta of the white rats was studied. At the 15th and 20th day of pregnancy the animals were killed by decapitation under ether narcosis. Immunochistochemical study revealed that in the three layers of the placenta the degree of expression of AE1\AE3 was reduced in the second and third trimester.

Expression of CD133 in a layer of giant trophoblastic and trophospongiotic layer cells and endothelial layer was reduced.

Study of distribution of the marker ki67 indicates on low proliferative activity, the exception are the blood vessels that are involved in formation of vessels of the umbilical cord.

THE HEPATOPROTECTIVE EFFICACY OF CORVITIN AND ACETYLCISTEIN IN MOUSE MODEL OF ACUTE HEPATITIS

The aim of the study was assessment of the effectiveness of corvitin and acetylcistain, as well as of combination of above named drugs in mouse model of paracetamol (acetaminophen) induced acute hepatitis. Experiments were carried out on white, male mice (n=49) fitting established standard criteria. All investigational drugs in experimental groups of animals were administered 1,5 h before induction of hepatitis and after its creation by paracetamol (750mg/kg i/p) during 3 days by a single dose. In the control group of animals saline solution was administered in equivalent quantity. The efficacy of used drugs were determined by the following criteria: mortality, level of biochemical markers of hepatocytes cytolysis (ALT,AST) and liver mass. Obtained results were statistically processed.

In acute hepatitis it was shown profound alteration of the level of hepatic biochemical markers and liver mass that accompanied by high mortality. Hepatoprotective activity of corvitine and acetylcistain in case of their separate administration was similar. Combined administration of the drugs has shown the greatest hepatoprotective efficacy. Presumably corvitin prodused preventive and curative effects due to its antioxidative properties, while hepatoprotective effect of acetylcisteine was revealed by its antioxidative and SH-groups donator properties.

It was suggested that the combination of corvitin and acetylcistein would become in near future an effective remedy for correction of hepatic function during its severe damages and reduction of mortalitis.

The aim of the study was investigation of citrus peel from the polimexilatedflavans and mineral origin bioligical active compounds complex  Geominis on the redox balance of the intact animals’ blood.

The obtained results show, that citrus polimetoxilated flavonoidsinduced oxygen free radicals  nutrolisation and thus contribute to the decreaseof SOD activity.At the same time, a compensatory increase in catalase activity was detected, which may be caused decrease of the supressive effect of superoxides on this enzime activity. Free NO content in the blood did not statistically important chnged under the influence of the cytrus  polimetoxilated flavonoids.

Geomin did not influence the intact animal blood antioxidant enzymes, superoxide dismutase and catalase, activity and free NO-’s content.

EFFICACY OF CORVITIN AND L-CARNITIN IN MOUSE MODEL OF RAY SYNDROME

Efficacy of corvitin and L-carnitin, along with their combination was studied in the mouse model of Ray syndrome induced according with A.Vengersky’s method. Intraperitoneal administration of 4-pentenoic acid was performed once daily during seven days, the used dosage was 20mg/kg. The efficacy of drugs was assessed by the means of mortality data, biochemical markers of hepatocytes cytolysis (ALT, AST) and liver mass.

Performed investigation revealed that administration of 4-pentenoic acid elevates biochemical markers of hepatocytes cytolysis (ALT, AST) and increases mortality of experimental animals, without considerable change of liver mass.

Recently, the physiological role of the growth hormone (GH) in adult life has received much attention. According to data, received from numerous studies, GH has an important role, regarding the lipid and carbohydrate metabolism [1,2].

GH replacement therapy is crucially important for the adults with Hypopituitarism, which are highly vulnerable to increased vascular mortality and morbidity [3,4]. Those issues are the consequences of the dyslipidemia, which is seen in the patients with growth hormone deficiency.

GH replacement therapy in hypopituitary adults seems to produce beneficial effects on the plasma lipid profile. Therefore, has a huge importance in metabolic and weight regulation.

DIFFERENT ASPECTS OF COGNITIVE FUNCTIONING AND CASE OF AUTISM IN CHILDREN WITH FETAL ANTIEPILEPTIC DRUG EXPOSURE

It is well known that fetal antiepileptic drugs (AED) exposure is associated with an increased risk of congenital malformations, cognitive impairment and autistic spectrum disorders.

The aim of the study was to investigate AED exposure influence on different aspects of cognitive functioning  and risk of autistic spectrum disorders in children exposed in utero AED.

METHODS: 100 children from 3 to 6 year were participated in the study: 50 children of WWE received AED treatment during pregnancy and 50 children of women without epilepsy, control group (CG). All children underwent Wechsler Preschool and Primary Scale of Intelligence Test. Verbal and Non-verbal abilities, Full Scale IQ was calculate for each child.

RESULTS: The Verbal Comprehension Index was higher in children from CG group (mean – 97.06), compare to AED exposed children (84.4). Verbal concept formation, knowledge acquiredand verbal reasoning scores were significantly lower in children of  WWE compare to control group.

Visual Spatial Index was higher in unexposed individuals (mean – 101,5), compared to exposed children (86,4). Understanding part-whole relationships, organizing visual information, attending to visual detail and integrating visual and motor functions also were better in children from CG group.

The Full Scale IQ was significantly higher in children from CG group (101,5), compare to children of WWE (84.02).

When compare exposed children of WWE with different AED treatment, no significant difference revealed in cognitive functioning and Full Scale IQ scores.

CONCLUSION: In utero AED exposure may worsen child early cognitive development in general. It hinders development both, verbal and non-verbal cognitive abilities of preschool children.

Cardiovascular diseases present one of the major health problems worldwide. Flavonoids constitute the largest class of naturally occurring phenols of the plant origin, mostly existing in the free state or as their respective glycosides. They have received considerable attention due to their possible contribution to reducing risk for several chronic diseases.The present paper reviews the most recent studies aiming at evaluation of relationship of flavonoid intake with the risk of coronary heart disease.

THE EFFECTIVENESS OF CAMOUFLAGE IN THE COMPLEX TREATMENT OF PATIENTS WITH VITILIGÎ

Considering the impact of vitiligo on the psyche and self-esteem of the patient, the use of camouflage technology is very significant for the proper and global management of disease. To hide a cosmetic skin defect various cosmetic means are offered. It is now widely used tanning, coloring and bleaching lotions, liquid and compact base, fixing powder and sprays, semipermanent and permanent tattoo.

The aim of the study was to determine the effectiveness of camouflage in the complex treatment of patients with acrofacial vitiligo.

18-65 years old 35 patients with acrofacial form of vitiligo (20 women and 15 men) with disease duration from 1 to 15 years were under clinical observation. Vitiligo depigmented lesions were located on the face (around the eyes, and the mouth) on the upper limbs (dorsal surface of the hands with partial transition to the palms). All the patients received local UVB 311 phototherapy of vitiligo lesions  (portable device of Dr. Honle company). Patients underwent phototherapy sessions 3 times a week (treatment procedure envisaged 15 courses), observed patients were divided into 2 groups; main group and the comparison (control) group. According to data of different authors and results of own observations vitiligo lesions on the hands turned out to be the most difficult to treat. Based on this, the patients of main group for camouflage of lesions on the hands were using tonal pencil “ZANDERM”.  As we know vitiligo represents a significant cosmetic defect, which has an aggravating effect on the course of dermatosis and psychological conditions of patients

To study some psychological peculiarities observed in vitiligo patients before and after treatment (4 months) we used the “personal anxiety scale”. Both groups of patients before treatment have shown the high level of anxiety (25-40) points. After the combined treatment with phototherapy and the tonal pencil, the level of anxiety in the study (main) group was significantly reduced (5-15 points). While in comparison group they remained on the high level. Along with the positive dynamics of the level of anxiety in the main group, earlier appearance of repigmentation was observed on the face, and the repigmentation was homogenous without signs of desquamation and hyperkeratosis.

Thus, the complex treatment of patients with vitiligo by means of camouflage not only gives a positive cosmetic effect but also has positive influence on some of the psychological conditions of  vitiligo patients.

Tuberculosis data collected in the database of the NCTLD for 2005-2015 were analyzed. 453 presumptive genital tuberculosis (GTB) cases registered at the NCTLD during 2012-2014 were studied. High rates of GTB notification were observed in the capital and nearby districts. Out of 290 GTB cases registered at the NCTLD 16.2%  was already diagnosed in the different clinics through the morphology of operational material, while the GTB diagnoses at the NCTLD were verified by less invasive methods.

Rehabilitation influences of adaptive physical exercises are based on interrelation of impellent and vegetative functions. In particular, at corrective picked up physical exercises, according to functionalities of an organism, autonomic nervous activity causes positive changes in work of internal bodies.

Received data of researching and control group, allows conclude, that the complex adaptive exercises and correct by selected power of physical load, with the accounting of time, cause comparative rehabilitation autonomic nervous system for people with congenital aberration of genes, which is very important for their vital activity, from our point of view.

Use of animals in experiments have a long history, however modern biomedical research requires to provide proper care to all laboratory animals and respect ethical aspects based on internationally approved ethical guidelines and regulatory norms. There are no ethical standards and legal regulations of animal use in bio-medical research in Georgia. The aim of the study is to evaluate the attitude of the Georgian society towards animal use in biomedical research considering gender and age of respondents. Special questionnaire was elaborated. There’s no statistically significant difference between genders, however the differences were examined by age interval.

The structure of diseases in Georgia has changed dramatically - sharply increased the number of non-communicable diseases, mental disorders and drug addiction. It should be noted that Georgia’s health care system over the years was focusing on the technologies, which would require a sophisticated hospital care, high-tech operations and less attention was paid to disease prevention, early detection and their effective managment. Therefore special attention should be given to the access to the primary health care system and to increase its role, as in the countries where the medical services is oriented on the primary health care model, the satisfaction of the population is high, the prevention services are better developed, they are characterized by the better indicators of health and less health care expenses.

Was developed rapid, selective and effective Liquid Chromatography – Tandem Mass Spectrometry (LC-MS/MS) method for determinationof risperidone in human urine, quetiapine fumarate was used as internal standard.For isolation was used solid phase extraction with the cartridge Supelclean LC-18 SPE 500mg. In vitro urine samples were chromatographed on AGILENT TECHNOLOGIES 1290 Infinity AGILENT TECHNOLOGIES 6460 Triple quad LC-MS/MS system. Separation was performed by isocratic elution on Zorbax Eclipse plus C18 (100´4.5 mm, 3.8 µm) column. Determination was performed on a triple-quadrupole masspectrometer employing electrospray ionization technique (ESI+) operating in multiple reaction monitoring (MRM) mode. The optimized MRM transitions for risperidone were m/z 410® 191, m/z 410® 163 and m/z 410® 110. Calibration curve show a linear relationship in the range of 0.5 to 100 ng/ml.

SEIZURES AND EPILEPTIFORM EEG-DISCHARGES IN CHILDREN WITH FOETAL EXPOSURE OF ANTIEPILEPTIC DRUGS

It is well known that foetal AEDs exposure is associated with an increased risk of congenital malformations, cognitive impairment and autistic spectrum disorders.

The aim of the current study was to assess the effects of foetal AEDs exposure on the seizures and epileptiform EEG discharges in children.

A prospective cohort group of children aged 3-6 years (MD - 4.5 years) with fetal exposure to AEDs from mothers with epilepsy (Case-group) was formed. Age matched individuals without fetal AEDs exposure from healthy women were enrolled as a control group. In all of the 96 children from case (n=47) and control (n=49) groups standard EEG and neurological/epileptological examinations were performed.

Specific epileptiform EEG-discharges  and nonspecific EEG abnormalities  were observed in 21% of children from case group (10/47); in one child (2%) of control group specific epileptiform EEG discharges (spike and wave discharges), without clinical seizures (1/49) were revealed. Epilepsy was diagnosed in three children (6%) from case group: two of them were siblings with foetal VPA exposure and focal epilepsies were diagnosed in the both of them. In the remaining one child of the mother with reflex epilepsy and foetal VPA-exposure, reflex epilepsy was diagnosed.

Epileptiform EEG discharges are tenfold and the seizures are three times as often found in children with fetal AEDs exposure from mothers with epilepsy than in their healthy peers from healthy mothers. These effects may be due to epilepsy itself or foetal AEDs exposure may play some role that needs further research.

The research is to estimate of post-strategies and make relatively recommendation of stroke. It used to research clinic, 31 patient and also used it in acute phase, at last of hospitalization and conduced the methods of rehabilitation resort.

The research has shown, the basic role is that in period of post-stroke rehabilitation at the moment of diseases processing. So, trustworthy of prognosis is depend on the time of beginning rehabilitation.

The administration of the appetite stimulant for children with disorders of nutritional status was analyzed. It was found that the use of pediatric syrup of cyproheptadine in a short 2-week period improves the body weight parameters corresponding to the nutritional disturbance according to the criterion of “underweight”.
